Acidity-Sensitive Spacer Molecule To Control The Release Of Pharmaceuticals From Molecular Carriers

View page
US Patent:
46311904, Dec 23, 1986
Filed:
Jun 26, 1981
Appl. No.:
6/277633
Inventors:
Wei C. Shen - Needham MA
Hugues J. Ryser - Concord MA
International Classification:
A61K 3944
US Classification:
424 85
Abstract:
A method for controlling the release of drugs or other passenger moleculars from carrier conjugates, and a class of conjugates that releases drugs when ingested by a cell or subjected to acidic conditions, are disclosed. These conjugates contain a passenger molecule which is attached to a spacer molecule through an acidic bonding group, such as carboxyl, that is in a "cis" configuration with another acidic group, and a carrier molecule that is bonded to the spacer molecule at another site. When subjected to a mile increase in acidity, such as occurs within a lysosome of a cell, the drug or other passenger molecule is hydrolyzed from the conjugate and released in unmodified, active form.

US Classification:
5303915
Abstract:
A method for controlling the release of durgs or other passenger molecules form carrier conjugates, and a class of conjugates that releases drugs when ingested by a cell or subjected to acidic conditions, are disclosed. These conjugates contain a passenger molecule which is attached to a spacer molecule through an acidic bonding group, such as carboxyl, that is in a "cis" configuration with another acidic group, and a carrier molecule that is bonded to the spacer molecule at another site. When subjected to a mild increase in acidity, such as occurs within a lysosome of a cell, the drug or other passenger molecule is hydrolyzed from the the conjugate and released in unmodified, active form.

Method Of Effecting Cellular Uptake Of Molecules

View page
US Patent:
47015217, Oct 20, 1987
Filed:
Jan 10, 1979
Appl. No.:
6/002368
Inventors:
Hugues J. Ryser - Concord MA
Wei-Chiang Shen - Needham MA
Assignee:
The Trustees of Boston University - Boston MA
International Classification:
C07K 900
US Classification:
530322
Abstract:
A method of effecting cellular uptake of molecules which are either excluded from cells or poorly transported into cells is disclosed wherein such molecules are covalently bonded to a cationic polymer which serves as a transport carrier to transport the molecules into cells.
